Abstract  Finasteride is a specific inhibitor of type II 5alpha-reductase, the enzyme that converts testosterone (T) to the more potent androgen receptor agonist dihydrotestosterone (DHT). In utero exposure to androgen receptor antagonists and T biosynthesis inhibitors have induced permanent effects on androgen-sensitive end points such as anogenital distance (AGD), nipple retention, and malformations of the male rat reproductive tract. The objectives of this study were to (1) characterize the dose response of finasteride-mediated alterations in androgen-dependent developmental end points, (2) determine whether prenatal exposure to finasteride permanently decreases AGD or results in nipple retention, and (3) evaluate whether AGD or nipple retention is predictive of adverse alterations in the male reproductive tract. Pregnant Crl:CD(SD)BR rats (n=5-6/group) were gavaged with either vehicle or finasteride at 0.01, 0.1, 1.0, 10, or 100 mg/kg/day on gestation days 12 to 21. All male offspring were monitored individually until necropsy on postnatal day (PND) 90. The present study design has been used previously for other antiandrogens and is sensitive to perturbations of the male rat reproductive tract. Decreases in AGD on PND 1 and increases in areolae-nipple retention on PND 13 were significantly different from controls in all finasteride-exposed male rats. Finasteride-induced changes in AGD and nipple retention were permanent in male rats exposed to finasteride at and above 0.1 mg/kg/day. On PND 90, dorsolateral and ventral prostate lobes were absent in 21 to 24% of rats exposed to 100 mg/kg/day finasteride and weighed significantly less at and above 10 mg/kg/day. In the highest dose group, 73% of animals had ectopic testes, much higher than previously reported. The most sensitive malformation other than decreased AGD and nipple retention was the dose-dependent increase in hypospadias. The lowest observed adverse effect level (LOAEL) for finasteride-induced permanent effects in this study was 0.1 mg/kg/day based on permanent changes in AGD and nipple retention. Finasteride-induced changes in AGD and retention of nipples were highly predictive of hypospadias, ectopic testes, and prostate malformations even though some animals with retained nipples or decreased AGD may not have had other reproductive tract malformations. In summary, prenatal exposure to finasteride specifically inhibited DHT-mediated development with little to no change in T-mediated development.

Abstract  The present study describes atypical Leydig cell (LC) hyperplasia in 20-week-old Sprague-Dawley rats with low testosterone and high luteinizing hormone levels after prenatal administration of 100 mg/kg/day di(n-butyl) phthalate on days 12 to 21 postconception. Light microscopy revealed LC hyperplasia surrounded by severely degenerated seminiferous tubules. Aggregated LCs had large ovoid nuclei with nucleoli and abundant eosinophilic cytoplasm. Immunohistochemical analysis showed expression of proliferating cell nuclear antigen and vimentin in many hyperplastic LCs. Electron microscopy revealed atypical nuclei, abundant free ribosomes, stripped rough endoplasmic reticulum, intermediate-size filaments, elongated cytoplasmic filopodia, atypical tight junctions, and cilia formations, but smooth endoplasmic reticulum was scarcely observed.

Abstract  This book deals with the safe and legal handling of hazardous materials and waste from the manufacturer's plant through the storage, transportation and distribution channels to the user, and, ultimately, to the disposal of the product or waste materials. There is increasing pressure today from the public, academia, government at all levels, and industry to improve the handling and management of hazardous materials. A knowledge of the methods required to safely handle and manage those materials in all of their various aspects, together with an understanding of the many governmental regulations that apply to those materials in the various stages of the distribution chain, is absolutely essential to their proper handling and disposal. Efficient handling and the safe management of hazardous materials requires an expertise in the skills and techniques of the latest innovations, which in turn are often based upon the firm foundation of data and experience in those areas. Personal and public safety require that the information concerning hazardous materials be disseminated as widely as possible. This document should not be used to determine compliance with the u.s. DOT hazardous material regulations, or with any other regulations imposed by loca\, State, or Federal regulatory bodies.

Abstract  Il y a environ 50 000 à 60 000 composés chimiques utilisés communément à des fins techniques. Certains d'entre eux sont toxiques et leur rejet dans l'environnement peut constituer une menace pour l'équilibre des écosystèmes aquatiques et pour la santé humaine. Plusieurs listes de substances dangereuses, appelées également polluants chimiques prioritaires, ont ainsi été établies par des organismes nationaux ou internationaux, notamment par l'EPA aux Etats-Unis, la CEE en Europe, l'Organisation Mondiale de la Santé (OMS). La nécessité de protéger les milieux aquatiques et la santé humaine a par ailleurs amené le développement de standards, de critères de qualité des eaux naturelles et des eaux potables, vis-à-vis de ces contaminants chimiques. Les sources de pollution sont diverses, se partageant entre les sources localisées comme les effluents urbains et industriels et les sources diffuses comme les eaux de ruissellement en zones rurales et en zones urbaines et les retombées atmosphériques. Différentes processus biogéochimiques déterminent les mécanismes de transport et de transformation des polluants organiques dans le milieu aquatique, et la bioaccumutation dans les organismes vivants représente un important aspect de ce comportement. Une première évaluation de la qualité des eaux continentales (rivières, lacs, eaux souterraines) est effectuée en examinant les données recueillies à travers le réseau de surveillance mondiale de la qualité de l'eau (GENS/EAUX) mis en place par deux organismes des Nations Unies, l'OMS et le PNUE. D'autres sources d'informations, publiées dans la littérature scientifique, ont été également utilisées pour évaluer les niveaux de concentrations observés pour différents groupes de polluants organiques dans les eaux naturelles et les eaux potables. La plupart des données disponibles proviennent d'un nombre limité de pays industriels d'Amérique du Nord, d'Europe et du Japon. Il y a une dramatique absence d'informations concernant la qualité chimique des eaux continentales dans les pays les moins développés d'Afrique, d'Amérique Latine et d'Asie. A l'heure actuelle, la stratégie de surveillance des polluants chimiques à l'échelle mondiale est donc loin d'être réellement satisfaisante.

Abstract  The marine environment provides a sink for a host of toxic chemicals, directly or inadvertently, released as a result of human activity. Some of these chemicals have the potential to act as aneugens, substances that cause numerical chromosomal aberrations (NCAs). NCAs are one of the most important classes of genetic abnormality and are implicated in a variety of deleterious effects, including premature ageing, birth defects and cancer. Clearly, any increase in the incidence of these agents in the marine environment poses a risk to the indigenous biota and its predators, including man. In this paper, we describe our recent success with applying the fluorescence in situ hybridisation technique (FISH) to detect NCAs in the interphase cell nuclei of Pomatoceros lamarckii, a common rocky shore invertebrate. Given the lack of requirement for any detailed cytogenetic knowledge, the method holds considerable promise for laboratory and field studies in general, and should lend itself to automated screening protocols, where large numbers of cells can be screened rapidly, for example, using a flow cytometer. When exposed either under acute or chronic (viz. adult) exposure conditions, colchicine and di-butylphthalate (DBP) (a widely-used plasticiser), two recognised aneugens, induced significant increases in the levels of NCAs, in the dose range 1 x 10(-6)-5 x 10(-6) M, in both four to eight cell embryo stages and 24 h-old larvae. In keeping with the severely debilitating effects of this class of agent, an inverse correlation was observed between the induced levels of NCAs and larval fitness based on the results of a standard 48-h larval bioassay.

Abstract  To study the surface structure of MgCl(2) support and its interaction with other active components in Ziegler-Natta catalyst, such as electron donors, we prepared a thin film analogue for Ziegler-Natta ethylene polymerization catalyst support by spin-coating a solution of MgCl(2) in ethanol, optionally containing a diester internal donor (diisobutyl-ortho-phthalate, DIBP) on a flat Si crystal surface. The donor content of these films was quantified by applying attenuated total internal reflection-Fourier transform infrared spectroscopy (ATR-FTIR) and X-ray photoelectron spectroscopy (XPS). Changes in the interaction of DIBP with MgCl(2) at various temperatures were monitored by in situ ATR-FTIR. Upon increasing the temperature, a shift in the (C═O) band toward lower wavenumbers was observed together with the depletion of (O-H) stretching band due to the desorption of residual ethanol. We assign this shift to gradual redistribution of adsorbed DIBP from adsorption sites on the MgCl(2) (104) surface toward the more acidic MgCl(2) (110) surface. The morphologies of MgCl(2) and MgCl(2)/DIBP films were studied by transmission electron microscopy (TEM) revealing a preferential orientation of ClMgCl layers (001) parallel to the lateral film dimensions. This orientation becomes more pronounced upon annealing. In the absence of donor, the MgCl(2) grow in to large crystals aligned in large domains upon annealing. Both crystal growth and alignment is impeded by the presence of donor.

Abstract  Abstract  Polyvinyl chloride (PVC) flooring material contains phthalates, and it has been shown that such materials are important sources for phthalates in indoor dust. Phthalates are suspected endocrine-disrupting chemicals (EDCs). Consecutive infants between 2 and 6 months old and their mothers were invited. A questionnaire about indoor environmental factors and family lifestyle was used. Urinary metabolites of the phthalates diethyl phthalate (DEP), dibutyl phthalate (DBP), butylbenzyl phthalate (BBzP), and dietylhexyl phthalate (DEHP) were measured in the urine of the children. Of 209 invited children, 110 (52%) participated. Urine samples were obtained from 83 of these. Urine levels of the BBzP metabolite monobenzyl phthalate (MBzP) was significantly higher in infants with PVC flooring in their bedrooms (P < 0.007) and related to the body area of the infant. Levels of the DEHP metabolites MEHHP (P < 0.01) and MEOHP (P < 0.04) were higher in the 2-month-old infants who were not exclusively breast-fed when compared with breast-fed children. The findings indicate that the use of soft PVC as flooring material may increase the human uptake of phthalates in infants. Urinary levels of phthalate metabolites during early life are associated with the use of PVC flooring in the bedroom, body area, and the use of infant formula. PRACTICAL IMPLICATIONS: This study shows that the uptake of phthalates is not only related to oral uptake from, for example, food but also to environmental factors such as building materials. This new information should be considered when designing indoor environment, especially for children.

Abstract  In metabolomic studies using liquid chromatography mass spectrometry of urine from children with cystic fibrosis (CF), high levels of metabolites of low molecular weight phthalates were found. Phthalate metabolite excretion was explained by therapy with enteric-coated pancreatic enzyme replacements. Phthalate metabolite identity was confirmed by tandem mass spectrometry. Pancreatic insufficient CF children taking Cotazym ECS(®), which is formulated with diethyl phthalate (DEP), had urinary metabolites of DEP. Children taking Creon(®), which has dibutyl phthalate (DBP), excreted DBP metabolites. The estimated concentrations of free MEP were 2-3 orders of magnitude higher than reported from environmental phthalate exposure. Enteric-coated pancreatic enzymes can expose individuals with CF to incessant, high oral intakes of phthalates. Although adverse effects have neither been shown to be present nor absent, we raise the need to consider that individuals requiring life-long therapy with some current pancreatic enzyme replacements chronically ingest high amounts of phthalates.

Abstract  Thermoreversible gelation of poly(vinylidene fluoride) (PVDF) has been studied in a new series of solvents (phthalates), for example, dimethyl phthalate (DMP), diethyl phthalate (DEP), dibutyl phthalate (DBP), and dihexyl phthalate (DHP) as a function of temperature and polymer concentration, both by test tube tilting and dynamic light scattering (DLS) method. The effect of aliphatic chain length (n) of diesters on the gelation kinetics, structure/microstructure and morphology of PVDF gels has been examined. Gelation rate was found to increase with increasing aliphatic chain length of diester. DLS results indicate that the sol-gel transformation proceeds via two-steps: first, microgel domains were formed, and then the infinite three-dimensional (3D) network is established by connecting microgels through polymer chains. The crystallites are responsible for 3D network for gelation in phthalates, and alpha-polymorph is formed during gelation producing higher amount of crystallinity with increasing aliphatic chain length of diester. Morphology of the networks of dried gels in different phthalates showed that fibril thickness and lateral dimensions decrease with higher homologues of phthalates. The scattering intensity is fitted with Debye-Bueche model in small-angle neutron scattering and suggested that both the correlation length and interlamellar spacing increases with n. A model has been proposed, based on electronic structure calculations, to explain the conformation of PVDF chain in presence of various phthalates and their complexes, which offer the cause of higher gelation rate for longer aliphatic chain length.

Abstract  A fast and simple method, which involves dynamic liquid-phase microextraction followed by gas chromatography with flame ionization detection, has been developed to facilitate the identification and quantification of three phthalate esters (dimethyl phthalate (DMP), diethyl phthalate (DEP), and di-n-butyl phthalate (DnBP)) in water samples. Experimental parameters affecting the extraction efficiency, such as extraction solvent, solvent volume, dwell time and extraction frequency were investigated. Under the optimal conditions, the method yields a linear calibration curve over three orders of magnitude for the target analytes. The limits of detection of DMP, DEP and DnBP were 4.30, 0.66 and 0.43 ng mL(-1), respectively, and relative standard deviations at the 100 ng mL(-1) level were 6.4%, 5.4% and 5.2%, respectively (n=6). Lake water, tap water and bottled mineral water samples were successfully analyzed using the proposed method.

Abstract  A nearly symmetric polystyrene-block-polyisoprene diblock copolymer dissolved at a concentration of 40% in styrene-selective solvents exhibited a cylinder-to-disorder transition upon heating. The solvents used were diethyl phthalate (DEP) and 75:25 and 50:50 mixtures of DEP with di-n-butyl phthalate (DBP). In DEP, the most styrene-selective of the three solvents, rheological measurements indicated a distinct plateau in the temperature-dependent elastic modulus across the 8 degrees C interval above the order-disorder transition temperature, T(ODT) = 116 degrees C. Previous small-angle neutron scattering measurements in this regime indicated the equilibrium phase to be a liquidlike solution of approximately spherical micelles. An isothermal frequency sweep in this regime indicated a very long relaxation time. Annealing eventually led to the recovery of liquidlike rheological response, over a time scale of hours. Qualitatively similar phenomena were also observed in 75:25 DEP/DBP and 50:50 DEP/DBP solutions, except the fact that the temperature window of the transient response is narrow and the time scale for the recovery diminishes significantly. Neither small-angle X-ray scattering nor static birefringence gave any clear signature of the transient structure. The structure that leads to the transient rheological response is attributed to micellar congestion due to the slow relaxation of anisotropic micelles into an equilibrium distribution of micelles. Possible origins of the remarkable solvent selectivity dependence are also discussed.

Abstract  Phthalates (endocrine disrupters) released from food-contacted plastics into aqueous solution during microwave conditioning were measured with microdialysis enrichment on-lined high-performance liquid chromatography. The released phthalates in aqueous solution were diffused through a cellular dialysis membrane into the perfusion stream and thus enriched prior to HPLC analysis. Conditions for obtaining optimum enrichment such as the hollow dialysis fiber, flow-rate and polarity modifier in perfusion stream, pH, added-salt and stirring rate in sample solution, as well as chromatographic conditions were investigated. Experimental results indicated that microdialysis enrichment with a 20-cm polysulfone hollow dialysis fiber and heptane as the perfusate at 0.10-microL/min flow-rate to collect phthalates from aqueous sample in 0.5M KCl matrix (optional pH) at 250 rpm stirring offered the optimum enriched efficiency. The dimethyl phthalate (DMP), diethyl phthalate (DEP) and dibutyl phthalate (DBP) were well separated within 16 min by a C-18 column and eluted gradient from 40 to 90% aqueous acetonitrile (at pH 6.0) and 1.0 to 1.5 mL/min flow-rate. Detection was carried out with an UV detector at 225 nm. The enrichment factors were 14, 140 and 201 (at 0.10-microL/min perfusate flow-rate) for DMP, DEP and DBP, respectively, with less than 4% RSD. The proposed method provided a very simple, fast and eco-friendly enriched procedure to determine the extent of phthalates migration from disposable plastic materials into drinking soup.

Abstract  Prochloraz (PZ) is an imidazole fungicide that displays multiple endocrine activities. It inhibits steroid synthesis via P450 modulation and acts as an androgen receptor (AR) antagonist, but its effects on male sexual differentiation have not been described. The purpose of the current study was to expand in vitro observations and to determine whether PZ affected sexual differentiation. PZ effects on AR-mediated gene expression were tested using a cell line (MDA-kb2) containing endogenous AR and stably transfected with an MMTV-luc reporter. PZ concentrations greater than 1 microM caused a dose-dependent inhibition of dihydrotestosterone-induced gene expression. PZ also inhibited R1881 binding to the rat AR (IC50 approximately 60 microM). In vivo, pregnant rats received PZ by gavage from Gestational Day 14 to 18 at doses of 31.25, 62.5, 125, and 250 mg/kg of body weight per day. PZ delayed delivery in a dose-dependent manner and resulted in pup mortalities at the two highest doses. In male offspring, anogenital distance and body weight were slightly reduced at 3 days of age. Additionally, female-like areolas were observed at 13 days of age at frequencies of 31%, 43%, 41%, and 71% in the lowest-dose to highest-dose groups, respectively. Weights of androgen-dependent tissues showed dose-dependent reductions. Hypospadias and vaginal pouches were noted in all males treated with 250 mg/kg, whereas those defects were observed in 12.5% and 6.25%, respectively, of males treated with 125 mg/kg. Treatment did not affect age of preputial separation in animals without penile malformations. Despite severe malformations in males, no malformations were noted in females. Together, these results indicate that PZ alters sexual differentiation in an antiandrogenic manner.

Abstract  Ready-to-eat packed meals intended to hospital patients were studied over a two-weeks period to measure the contents of di-(2-ethylhexyl) phthalate (DEHP) and di-n-butylphthalate (DBP) and to evaluate their daily intake by total diet. The packaging consisted of polyethylene terephthalate (PET) dishes sealed with polypropylene (PP) foil. The DEHP mean concentrations in total meals varied from 0.061±0.028 to 0.307±0.138μg/gwetweight (wet wt.); the DBP mean levels varied from 0.025±0.018 to 0.174±0.091μg/gwetwt. Highest levels of concentration for DEHP and DBP were found in bread with mean values of 0.307±0.138μg/gwetwt. and 0.174±0.091μg/gwetwt. for DEHP and DBP, respectively. The daily intake for DEHP was 3.1±0.9μg/kgbw and 1.5±0.5μg/kgbw for DBP. The mean±sd incidence of DEHP and DBP intake via hospital meals on the respective EFSA TDI was 6±2% (range 4-11%), and 15±5% (range 8-24%), respectively. Even if for hospital patients the major route of exposure may be represented by medical devices, the influence of the diet could have a significant value on TDI.

Abstract  In our effort to develop novel plant-derived acaricides, we examined the contact and fumigant toxicity of Asarum heterotropoides (Asarum sieboldii Miquel) essential oil constituents to the house dust mite Dermatophagoides farinae (Acari: Pyroglyphidae). Ten constituents, including methyl eugenol (relative amount 42.18 %), were identified by gas chromatography-mass spectroscopy (GC-MS) in the A. sieboldii Miq. essential oil. In contact toxicity tests, methyl eugenol (4.2 μg/cm(2), 24 h LD50) was most toxic to D. farinae, followed by benzyl benzoate (9.1 μg/cm(2)), A. sieboldii Miq. essential oil (37.7 μg/cm(2)), and dibutyl phthalate (DBP 57.9 μg/cm(2)). The potency of methyl eugenol and A. sieboldii Miq. essential oil was higher than benzyl benzoate and DBP, with mortalities of 100, 100, 94.6, and 13.2 %, respectively, after 2.5 h of exposure. In the vapor phase mortality bioassay, methyl eugenol and A. sieboldii Miq. essential oil resulted in 100 % mortality in closed containers after 24-h exposure, but only 4.7 and 7.9 %, respectively, in open containers, indicating that the toxicity in these tests was largely due to the vapor phase. Methyl eugenol and A. sieboldii Miq. essential oil merit further study as potential D. farinae control compounds.

Abstract  A dibutyl phthalate (DBP) transforming bacterium, strain M673, was isolated and identified as Acinetobacter sp. This strain could not grow on dialkyl phthalates, including dimethyl, diethyl, dipropyl, dibutyl, dipentyl, dihexyl, di(2-ethylhexyl), di-n-octyl, and dinonyl phthalate, but suspensions of cells could transform these compounds to phthalate via corresponding monoalkyl phthalates. During growth in Luria-Bertani medium, M673 produced the high amounts of non-DBP-induced intracellular hydrolase in the stationary phase. One DBP hydrolase gene containing an open reading frame of 1,095 bp was screened from a genomic library, and its expression product hydrolyzed various dialkyl phthalates to the corresponding monoalkyl phthalates.

Abstract  Reproductive disorders that are common/increasing in prevalence in human males may arise because of deficient androgen production/action during a fetal 'masculinization programming window'. We identify a potentially important role for Chicken Ovalbumin Upstream Promoter-Transcription Factor II (COUP-TFII) in Leydig cell (LC) steroidogenesis that may partly explain this. In rats, fetal LC size and intratesticular testosterone (ITT) increased ~3-fold between e15.5-e21.5 which associated with a progressive decrease in the percentage of LC expressing COUP-TFII. Exposure of fetuses to dibutyl phthalate (DBP), which induces masculinization disorders, dose-dependently prevented the age-related decrease in LC COUP-TFII expression and the normal increases in LC size and ITT. We show that nuclear COUP-TFII expression in fetal rat LC relates inversely to LC expression of steroidogenic factor-1 (SF-1)-dependent genes (StAR, Cyp11a1, Cyp17a1) with overlapping binding sites for SF-1 and COUP-TFII in their promoter regions, but does not affect an SF-1 dependent LC gene (3β-HSD) without overlapping sites. We also show that once COUP-TFII expression in LC has switched off, it is re-induced by DBP exposure, coincident with suppression of ITT. Furthermore, other treatments that reduce fetal ITT in rats (dexamethasone, diethylstilbestrol (DES)) also maintain/induce LC nuclear expression of COUP-TFII. In contrast to rats, in mice DBP neither causes persistence of fetal LC COUP-TFII nor reduces ITT, whereas DES-exposure of mice maintains COUP-TFII expression in fetal LC and decreases ITT, as in rats. These findings suggest that lifting of repression by COUP-TFII may be an important mechanism that promotes increased testosterone production by fetal LC to drive masculinization. As we also show an age-related decline in expression of COUP-TFII in human fetal LC, this mechanism may also be functional in humans, and its susceptibility to disruption by environmental chemicals, stress and pregnancy hormones could explain the origin of some human male reproductive disorders.

Abstract  Atmospheric trace phthalic esters (PAEs) carried by total suspended particulates were systematically investigated. A total of 450 air samples were collected at six typical locations and three specific micro-environmental sites (a stadium with a new plastic track, new cars, and a farming greenhouse with plastic film) in Nanjing metropolitan area of China from April 2009 to January 2010. The samples were analyzed by high-performance liquid chromatography and ultraviolet detection. The results are summarized as follows: (1) The dominant PAEs are dimethyl phthalate (DMP), dibutyl phthalate (DBP), and diethylhexyl phthalate (DEHP), which were found in the atmosphere of Nanjing. The average concentrations of DMP, DBP, and DEHP were 10.5 ± 1.2, 62.3 ± 4.5, and 33.3 ± 2.5 ng m(-3), respectively, constituting 9.9%, 58.7%, and 31.4% of total PAEs (106 ± 8.2 ng m(-3)). (2) The dynamic variations of atmospheric PAEs in the three specific micro-environmental sites showed that the PAE concentrations in the stadium with a new plastic track reduced to normal after 1 month remediation, while new cars need 6 months to remove their effect. The levels of PAEs in a farming greenhouse with plastic film were relatively high, but little PAEs accumulated in vegetables. (3) The vertical profiles from 1.5 to 40 m above ground display some fluctuations in PAE concentration, but no significant height dependence. This information will make a valuable contribution to the examination of the influence of atmospheric PAEs on the environment and human health.

Abstract  OBJECTIVES: Although phthalates like dibutyl phthalate (DBP) and di-2-ethylhexyl phthalate (DEHP) are commonly used as plasticizers and their metabolites are especially suspected of reproductive toxicity, little is known about occupational exposure to those phthalates. The aim of this study was to assess the utility of measuring the metabolite concentrations of DBP and DEHP in serum and urine samples as an indicator of occupational exposure to those phthalates.

METHODS: Phthalate metabolites were analyzed by using column-switching high-performance liquid chromatography tandem mass spectrometry (LC-MS/MS).

RESULTS: We detected phthalate metabolites in serum and urine matrices at approximately 10-fold lower than the limit of detection of those metabolites in the same matrix by LC-MS/MS without column switching, which was sufficient to evaluate concentrations of phthalate metabolites for industrial workers and the general population.

CONCLUSION: The accuracy and precision of the analytical method indicate that urinary metabolite determination can be a more acceptable biomarker for studying phthalate exposure and adverse health outcomes.

Abstract  Accurate simultaneous analysis of different anionic species using ion-selective electrodes (ISEs) can be achieved even for non-specific sensors by resorting to an ordinary least squares multiple regression in the vicinity of the predicted concentrations. In this work the potentialities of this approach are evidenced by the determination of nitrate and chloride in synthetic and real water samples in which chloride concentration was significantly higher than nitrate. An AgCl/Ag(2)S electrode based on a homogeneous crystalline membrane together with a PVC electrode based on tert-octylammonium bromide dissolved in dibutylphthalate were used as potentiometric detectors for chloride and nitrate, respectively. For the implementation of the procedure, an automatic system based on sequential injection analysis was used. The results obtained by the standard addition method were biased for low concentrations of nitrate and were dependant on the relative proportion of NO(3)(-)/Cl(-). The results obtained by the proposed methodology for chloride determination were slightly better when compared to those obtained by the standard addition method. In relation to nitrate determination, the proposed methodology yielded values with a relative root mean square error of prediction (RRMSEP) of 2.8%, while for standard addition calibration, mean error values were approximately 12.1%.

Abstract  A coated graphite-epoxy ion-selective electrode for bismuth(III), based on the ion-pair between the [Bi(EDTA)](-) anion and tricaprylylmethylammonium cation (Aliquat 336S) incorporated onto a poly(vinylchloride) (PVC) matrix is constructed. A thin membrane film of this ion-pair, dibutylphthalate (DBPh) or ortho-nitrophenyloctylether (o-NPOE) in PVC was deposited directly onto a Perspex(R) tube which contained a graphite-epoxy conductor substrate attached to the end of a glass tube. The coating solution was prepared by dissolving 30% (w/w) of PVC in 10 ml of tetrahydrofuran following addition of 65% (w/w) DBPh or o-NPOE and 5% (w/w) of the ionic pair. The effect of pH, EDTA concentration and some cation and anion on the electrode response is investigated. The bismuth(III) ion-selective electrode shows a linear response in the bismuth(III) concentration range from 1.0 x 10(-8) to 1.0 x 10(-1) mol 1(-1) and 1.0 x 10(-7) to 1.0 x 10(-1) mol 1(-1) and a slope of 56.8 and 59.2 mV dec.(-1) for the polymeric membranes containing DBPh and o-NPOE, respectively. The lifetime of this electrode was superior to 1 year (over 1600 determinations for each polymeric membrane), with practical detection limits of 6.3 x 10(-9) and 4.4 x 10(-8) mol 1(-1) with these plasticizers. Application of this electrode with bismuth(III) determination in a stomach anti-acid sample is described.

Abstract  The National Toxicology Program (NTP) Center for the Evaluation of Risks to Human Reproduction (CERHR) conducted an evaluation of the potential for di-n-butyl phthalate (DBP) to cause adverse effects on reproduction and development in humans. DBP is one of 7 phthalate chemicals evaluated by the NTP CERHR Phthalates Expert Panel. These phthalates were selected for evaluation because of high production volume, extent of human exposures, use in children's products, and/or published evidence of reproductive or developmental toxicity. Unlike many phthalates, DBP is not currently used as a plasticizer in polyvinyl chloride plastics. DBP is a component of latex adhesives and is used in cosmetics and other personal care products, as a plasticizer in cellulose plastics, and as a solvent for dyes. The results of this evaluation on DBP are published in a NTP-CERHR monograph which includes: 1) the NTP Brief, 2) the Expert Panel Report on the Reproductive and Developmental Toxicity of Di-n-Butyl Phthalate, and 3) public comments received on the Expert Panel Report. As stated in the NTP Brief, the NTP reached the following conclusions regarding the possible effects of exposure to DBP on human development and reproduction. First, although DBP could possibly affect human reproduction and development if exposures are sufficiently high, the NTP concludes that there is negligible concern for reproductive toxicity in exposed adults. Second, the NTP concludes that there is minimal concern for developmental effects when pregnant women are exposed to DBP levels estimated by the panel (2-10 mug/kg body weight/day). There is no direct evidence that exposure of people to DBP adversely affects reproduction or development, but studies reviewed by the expert panel show that oral exposure to high doses of DBP (>/=100 mg/kg body weight/day) may adversely affect the prenatal and early postnatal development in rodents. Finally, based on exposure estimates in women of reproductive age, the NTP concludes that there is some concern for DBP causing adverse effects to human development, particularly development of the male reproductive system.

Abstract  The new cyclic polyazacycloalkane 1.4,8,11-tetra(n-octyl)-1,4,8,11-tetraazacyclotetradecane (L1) was synthesised and the copper(II) complex [Cu(L1)]2+ characterised. Different electrodes were prepared using the [Cu(L1)]2+ complex as ionophore, PVC as plastic matrix and o-nitrophenyl octyl ether (NPOE), bis(2-ethylhexyl) sebacate (BEHS) or dibutyl phthalate (DBP) as plasticizers. The electrode containing DBP showed a Nernstian response over a wide pH range and a fast response time (ca. 3 s) whereas NPOE and BEHS gave near-Nernstian slopes. Selectivity coefficients for the different anions with respect to perchlorate were calculated. The response of the electrodes basically followed the Hofmeister sequence, suggesting that interaction of the ionophore with the anions is via electrostatic forces rather than due to anion coordination to the axial sites of the square-planar [Cu(L1)]2+ complex.
