| Abstract | A modified in vitro system for the metabolism of 2-acetylaminofluorine (2-AAF) was developed. The system depends upon utilization of a NADPH-generating system, low amounts of microsomal protein (0.5-1.0 mg) and substrate (660 ng), and utilization of HEPES buffer. With these modifications, the in vitro metabolism of 2-AAF was found to be linear with time and after 30 min incubation, formation of N-hydroxy-2-acetylaminofluorine (N-OH-2-AAF) represented 30 % or greater of the 2-AAF added originally. Using this modified in vitro system in a 14-day preliminary study, mice fed a purina diet were found to metabolize 30% more of the added 2-AAF to N-OH-2-AAF than did mice receiving a synthetic diet. In another study, 2-AAF was added to diets of mice (100 ppm in Purina or synthetic) for periods of up to 18 weeks. Determinations of in vitro 2-AAF metabolism were made at periodic intervals and it was found that feeding of 2-AAF in the diet exerted little, if any influence on the conversion of 2-AAF to N-OH-2-AAF or to the 5- or 7-OH metabolites when compared to control animals. |
